Neuroprotective effect of lacosamide on cognitive dysfunction in Streptozotocin induced Alzheimer disease

Abstract Alzheimer disease (AD) is a chronic and progressive neurodegenerative disorder characterized by memory loss cognition impairment. A link has been established between AD epilepsy sharing multiple mechanisms pathogenesis. Similar Hippocampal changes have found both diseases. Choosing antiepileptic drug in patient represent great challenge especially with increase seizure risk patients. Lacosamide, drug, was reported to neuroprotective effect animal models histone deacetylase inhibition activity. This study designed investigate the potential of lacosamide treatment Streptozotocin induced male Wistar rats. Methods model single bilateral intracerebroventricular injection STZ (3 mg/kg) on day one. Lacosamide (30 mg/kg orally, once daily) administrated for 21 days. Cognitive function assessment done Morris Water Maze (MWM) Y tasks. APP MAPT mRNA level were measured. Results ICV-STZ caused significant prolongation Escape latency time reduction spent target quadrant MWM spontaneous alteration ratio compared control group. Up-regulation Amyloid precursor protein Microtubule associated tau gene expression. Chronic attenuated cognitive impairment mitigated expression STZ. Conclusion against deficits ameliorating Aβ Tau pathology.
